The parasitoids of Parlatoria ziziphi (Lucas) (Hemiptera: Diaspididae): with descriptions of two new species of Aphytis (Hymenoptera: Aphelinidae) from China, and a note on Aphytis species with black-tipped mid tibiae in the male SCIE

Abstract :

During investigations in the Fujian and Hunan Provinces of China, 5 parasitoids associated with Parlatoria ziziphi were identified based on morphological characteristics and 28SrDNA sequences. They comprised 4 ectoparasitoids: A. chrysomphali (Mercet), Aphytis jinshanensis Wang & Huang, sp.n., Aphytis nigromaculata Wang & Huang, sp.n., and A. nr. melinus DeBach, and 1 endoparasitoid, Encarsia citrina (Craw). The 2 new species are described and illustrated in detail, and the relationships of the 4 Aphytis parasitoids treated here are analyzed, resulting in a phylogenetic tree based on 28SrDNA sequences. Moreover, the 5 Aphytis species (including 1 new species described in this work) having the mid-tibia tipped with black in the male are summarized with their scale hosts and distribution, and an identification key to these species is provided.

夜蛾黑卵蜂是重大入侵害虫草地贪夜蛾的重要卵期寄生蜂。为明确夜蛾黑卵蜂的种内识别及过寄生现象,测定了夜蛾黑卵蜂对被同种自体(同一个体)或异体(不同个体)雌蜂寄生不同时间间隔后的寄主卵的识别能力,并研究了单头和多头雌蜂在不同寄主密度下的过寄生率。结果发现,无论自体或异体寄生识别,夜蛾黑卵蜂的过寄生率均为0。单头雌蜂在不同蜂卵比下寄生24 h后,寄主卵内寄生蜂1龄幼虫的百分比均小于100%,且每粒寄主卵内只有1头1龄幼虫。多头雌蜂在寄主数量充足条件下,当蜂卵比为1:100、2:200、4:400、6:600、8:800时,未被寄生的寄主卵的百分比分别为52.8%、67.2%、63.2%、53.6%和51.2%,含1头寄生蜂1龄幼虫的寄主卵的百分比分别为47.2%、32.8%、36.8%、46.4%和48.8%;多头雌蜂在寄主数量不足条件下,当蜂卵比为2:20、3:21、4:20、7:21、20:20时,夜蛾黑卵蜂的寄生率分别为60.0%、83.0%、95.0%、94.0%和96.0%,过寄生率分别为11.0%、38.0%、64.0%、73.0%和92.0%;单粒寄主卵内寄生蜂1龄幼虫的平均数量分别为0.4、0.6、1.1、1.3和2.1头,最大数量分别为3.0、4.0、5.0、6.0和12.0头。结果表明,夜蛾黑卵蜂具有自体和异体寄生识别能力;单头雌蜂的过寄生率与寄主数量无关;多头雌蜂在寄主数量充足的条件下,不会产生过寄生;但在寄主数量不足的条件下,随着蜂卵比升高,寄生率、过寄生率、单粒草地贪夜蛾卵内寄生蜂1龄幼虫的平均数和最大数均随之增加。蜂卵比为4:20时寄生率高,过寄生率低,内部竞争低,是较为合适的接蜂比例。

Abstract :

Seventeen species of the mali-group of Aphelinus (Hymenoptera: Aphelinidae) are reviewed worldwide, including three new species, and five previously described species in China. The mali-group was defined by a combination of characters: head and body dark with parts of metasoma pale; fore wing with a complete row of setae inside the linea calva, or also only with a few setae in the angle between the linea and marginal vein; legs with mesocoxae, metacoxae and metatibia dark, metafemur pale. In this paper, three new species, Aphelinus tuberocephalus Wang & Huang, sp.n., Aphelinus hainanensis Wang & Huang, sp.n. and Aphelinus ruellia Wang & Huang, sp.n., are described and illustrated, with notes on one species, Aphelinus coreae Hopper & Woolley, new to China. A revised key to species of the mali-group is provided. Mitochondrial (COI) partial sequences were obtained successfully for A. tuberocephalus, A. hainanensis, A. ruellia and A. coreae.

堆蜡粉蚧寄生蜂种类鉴别

Abstract :

堆蜡粉蚧Nipaecoccus vastator (Maskell)[=Nipaecoccus viridis (Newstead)]是柑橘等果树和园林观赏植物的重要害虫。本文调查了福建省堆蜡粉蚧的寄生蜂资源;整理了世界上寄生堆蜡粉蚧的寄生蜂种类,包括初寄生蜂的跳小蜂科Encyrtidae 8属25种,一次重寄生蜂的跳小蜂科4属13种和蚜小蜂科Aphelinidae 1属1种,二次重寄生蜂的姬小蜂科Eulophidae 1属1种和棒小蜂科Signiphoridae 1属1种。分析了寄生蜂的种群及其与蚧寄主的食物链关系,首次明确堆蜡粉蚧的一次和二次重寄生蜂。同时,厘清了我国已知分布的堆蜡粉蚧寄生蜂8种,提出分种检索表,记述了主要寄生蜂种类的形态特征,并附形态特征照片,其中含1中国新记录种,麦长索跳小蜂Anagyrus mirzai Agarwal et Alam。

小黑瓢虫雌成虫滞育基因的转录组测序分析

Abstract :

为明确小黑瓢虫Delphastus catalinae滞育的分子机制,使用Illumina HiSeq2500高通量测序平台分析小黑瓢虫雌成虫非滞育期、滞育期和滞育解除期的相对转录水平,从转录组测序数据中选取碱性磷酸酶(alkaline phosphatase,ALP)基因、过氧化氢酶(catalase,CAT)基因、乳酸脱氢酶(lactate dehydrogenase,LDH)基因、过氧化物酶(peroxidase,POD)基因、超氧化物歧化酶(superoxide dismutase,SOD)基因、海藻糖酶(trehalase,TRE)基因、促葡萄糖转运1蛋白亚基基因Ref和SCoAL琥珀酰辅酶连接酶基因GDP-forming,采用实时荧光定量PCR(quantitative real-time PCR,qRTPCR)技术检测上述基因在小黑瓢虫雌成虫3个不同时期的表达特征。结果显示,从9个样品中共筛选出67.86 Gb的clean data;936 447条contig被组装成52 255条unigene,所有的unigene都通过BLAST对Nr数据库进行了注释,有23 539条unigene被匹配。通过对unigene进行COG、GO和KEGG分析,发现在非滞育组和滞育组之间以及滞育解除组和滞育组之间分别有3 690条和4 662条差异表达基因,其中571条差异表达基因在滞育期特异性表达,涉及116条KEGG富集通路。qRT-PCR验证了转录组测序的准确性,发现LDH基因在滞育期的表达量显著下降,而其余7个基因的表达量均上升。

基于两性生命表评价取食柑橘木虱对六斑月瓢虫和龟纹瓢虫生长发育及种群的影响

为有效防控重要害虫柑橘木虱Diaphorina citri,在实验室条件下测定取食柑橘木虱后六斑月瓢虫Cheilomenes sexmaculata和龟纹瓢虫Propylea japonica各虫态的发育历期,利用构建的年龄-阶段两性生命表对取食柑橘木虱后六斑月瓢虫、龟纹瓢虫生长发育及其种群参数进行研究。结果显示,取食柑橘木虱后六斑月瓢虫羽化前的发育历期为14.80 d,显著长于取食柑橘木虱后龟纹瓢虫的14.41 d;取食柑橘木虱后六斑月瓢虫和龟纹瓢虫产卵期分别为15.88 d和10.23 d,前者显著长于后者;取食柑橘木虱后六斑月瓢虫净增殖率、内禀增长率和周限增长率分别为25.48、0.10 d-1和1.10 d-1,取食柑橘木虱后龟纹瓢虫净增殖率、内禀增长率和周限增长率分别为14.13、0.06 d-1和1.06 d-1,均低于取食柑橘木虱后六斑月瓢虫;取食柑橘木虱后六斑月瓢虫和龟纹瓢虫的平均世代周期分别为33.87 d和44.95 d,前者显著短于后者。表明取食柑橘木虱后六斑月瓢虫和龟纹瓢虫均能完成整个世代,且六斑月瓢虫种群增长优于龟纹瓢虫,可大规模繁殖六斑月瓢虫,可用于柑橘木虱的生物防治。

Simple Summary Aphytis species (Hymenoptera: Aphelinidae) have been considered as the most important natural enemies in biological control of armoured scale insects (Hemiptera: Diaspididae). However, it is difficult to identify the species of the genus, particularly that of the A. lingnanensis group, based on adult morphological characters. Although the differences in pupal pigmentation of Aphytis species have been documented previously, they have not been much used as taxonomic characters for Aphytis species identification. In this study, we present four characteristic pigmentation patterns of Aphytis pupae, together with photographs, including the categories of: entirely yellow, partly dark brown, entirely or predominantly black, and partly black pupae. No significant intra-specific variation in pupal colour pattern was detected despite relatively high numbers of specimens examined, many from multiple, and different, origins. We summarize the present status of pupal pigmentation in the described species of Aphytis, according to the species groups, which could be used as an important supplementary diagnostic character for distinguishing species of Aphytis, especially closely-related species. Species identification of Aphytis on the basis of adult morphology is extremely difficult, especially in the A. lingnanensis group, with several cryptic species. Pupal pigmentation could be used as one of the taxonomic characters for Aphytis species, and in some instances, pupal pigmentation actually provided the first clue to the distinctness of cryptic Aphytis species. The present study investigated the full-grown larvae or younger pupae of Aphytis species, and pupal pigmentation and pupal development were observed and photographed. Four characteristic pigmentation patterns of Aphytis pupae were summarized including: entirely yellow, partly dark brown, entirely or predominantly black, and partly black. The species in the chilensis and mytilaspidis groups, and some unassigned species, generally have entirely, or predominantly and or partly black pupae. The species in the chrysomphali, funicularis, and proclia groups generally have the pupae entirely yellow. The species of the lingnanensis group have the pupae both entirely yellow, e.g., A. fisheri, and partly dark brown pupae, e.g., A. lingnanensis, A. holoxanthus and A. melinus. The pupae of Aphytis species in this study had a developmental duration of about 5-8 days at 27 +/- 1 degrees C, 70 +/- 5% RH and a photoperiod of 10L: 14D. It was found that the pupal skin was always melanized at the beginning stage, generally in the first day, among the pigmented pupae of Aphytis species. As development continued, the pigmentation became darker and the eye colour changed from pale red/brown to green. No significant intra-specific variation in pupal colour pattern was detected despite relatively high numbers of specimens examined, many from multiple, and different, origins. Overall, our study indicates that pupal pigmentation could be more helpful in species identification of Aphytis.

Bombyx mori Nucleopolyhedrovirus p26 Is Associated with Viral Late Stage Replication SCIE

Simple Summary p26 is conserved among all completely sequenced Lepidoptera baculoviruses, and some baculoviruses even have two copies of p26 (p26a and p26b), which suggested that p26 may have a basic role in the baculovirus infection cycle. p26 may be transcribed by the host RNA polymerase II in both early and late infection. Here, protein analyses showed that Bombyx mori nucleopolyhedrovirus (BmNPV) p26 levels were very low amounts during the early phases of infection, which then increased and then declined during the late infection phase. Thus, BmNPV p26 might have functions in both the cytoplasm and nucleus. Previous p26 knockout study indicated that p26 may be an auxiliary gene that does not influence key aspects of viral replication or transmission, and RNAi response to p26 may somewhat regulate viral replication. Therefore, in order to maintain low p26 expression and measure BmNPV p26 function, a RNAi-based knockdown method was chosen. The results indicated that high p26 expression during the middle interval is necessary for late-stage viral replication. Since p26 is not essential for baculovirus replication and transmission, it would be interesting to investigate whether p26 is involved in regulating host innate immune response. Bombyx mori nucleopolyhedrovirus (BmNPV) p26 is conserved among all Lepidoptera baculoviruses that have been completely sequenced thus far, and some baculoviruses even have two copies of p26, which suggested that p26 may play an important role in the virus infection cycle. This study aimed to characterize BmNPV p26. We found that BmNPV p26 transcripts were detectable as early as 3 h post-infection (hpi), and the transcript levels rapidly increased starting from 12 hpi. Western blot analysis using an anti-p26 polyclonal antibody demonstrated that the corresponding protein was also detectable from 6 hpi in BmNPV-infected cell lysates. Immunofluorescence analysis demonstrated that p26 was mainly dispersed in the infected cell cytoplasm, whereas the over-expressed fusion protein EGFP-p26 also accumulated in the nucleus. These results indicated that p26 is an early BmNPV gene and has functions both in the cytoplasm and the nucleus. RNAi-based knockdown of p26 could produce infectious virus and normal-appearing virions but decreased budded virus (BV) production in BmNPV-infected cells at 72 hpi. Moreover, the results of further quantitative PCR (Q-PCR) analysis indicated that the gp64 and p74 transcripts levels decreased significantly. These results indicated that BmNPV p26 may be associated with BmNPV replication during the late infection stage.

温度和光周期对小黑瓢虫滞育的调节作用 CSCD

在实验室条件下研究了温度(11℃、16℃、21℃、26℃和31℃)和光周期(16L:8D、14L:10D、12L:12D、10L:14D和8L:16D)对小黑瓢虫Delphastuscatalinae(Horn)滞育的影响。结果表明,温度、光周期及二者的相互作用均对小黑瓢虫滞育有显著影响。低温和短光照诱导成虫进入滞育。26℃和31℃时,测定的5个光周期均不能诱导滞育;而11℃、16℃和21℃时,滞育率随温度降低或光照时长缩短均呈升高的趋势。11℃和光周期8L:16D是诱导滞育的最适条件。11℃和光周期8L:16D诱导40d,成虫进入稳定的滞育状态,滞育率高达95.44%。同时发现,滞育成虫产卵前期随温度降低或光照时长缩短而延长。11℃和光周期8L:16D有利于维持滞育,延长成虫产卵前期。21℃和光周期14L:10D加速滞育的解除,缩短成虫产卵前期。

Abstract :

The Chinese species of the varius group of Coccophagus Westwood (Hymenoptera: Aphelinidae) are reviewed, including described species Coccophagus albifuniculatus (Huang), C. anchoroides (Huang), C. caudatus (Huang), C. dilatatus (Huang), C. equifuniculatus (Huang), C. fumadus Hayat (new record for China), C. lii (Huang), C. pellucidus (Huang), and one new species, Coccophagus yunnana Wang, Huang & Polaszek sp. nov. A revised key to the females of the Chinese species of the varius group of Coccophagus is provided. Mitochondrial (CO1) and nuclear ribosomal (28S-D2) partial sequences were obtained successfully for C. fumadus. http://www.zoobank.org/urn:lsid:zoobank.org:pub:264BBB77-9D32-4E9F-B31D-54E996FDEF7C
