The Effects of Composite Alkali-Stored Spent Hypsizygus marmoreus Substrate on Carcass Quality, Rumen Fermentation, and Rumen Microbial Diversity in Goats SCIE

Abstract :

The objective of this study was to investigate the effects of composite alkali-stored spent Hypsizygus marmoreus substrate (SHMS) on carcass quality, rumen fermentation, and rumen microbial diversity in goats. Twenty-four 6-month-old Chuanzhong black goats with similar body weights (20 +/- 5 kg) were selected and randomly divided into four groups (n = 6 per group) and received four treatments: 0% (control group, CG); 20% (low-addition group, LG); 30% (moderate-addition group, MG); and 40% (high-addition group, HG) of SHMS-replaced silage corn and oat hay. The experiment lasted for 74 days (including a 14 d adaptation period and a 60 d treatment period). The results of this study showed that MG and HG significantly improved the marble score of goat meat (p < 0.05). The flesh color score significantly increased in each group (p < 0.05). The fat color scores significantly increased in LG and MG (p < 0.05). There were no significant effects on the pH value or shear force of the longissimus dorsi in each group (p > 0.05). The cooking loss in MG was higher than that in CG (p < 0.05). The histidine and tyrosine contents in each group of muscles significantly increased (p < 0.05), with no significant effect on fatty acids (p > 0.05). The rumen pH of MG significantly decreased (p < 0.05), while the total volatile fatty acids (TVFAs) and ammoniacal nitrogen (NH3-N) increased by 44.63% and 54.50%, respectively. The addition of the SHMS altered both the alpha and beta diversities of the rumen microbiota and significant differences in the composition and structure of the four microbial communities. The dominant bacterial phylum in each group were Firmicutes and Bacteroidetes, with Prevotella 1 as the dominant bacterial genus. Correlation analysis revealed that rumen bacteria are closely related to the animal carcass quality and rumen fermentation. In the PICRUSt prediction, 21 significantly different pathways were found, and the correlation network showed a positive correlation between the Prevotella 1 and 7 metabolic pathways, while the C5-branched dibasic acid metabolism was positively correlated with nine bacteria. In summary, feeding goats with an SHMS diet can improve the carcass quality, promote rumen fermentation, and alter the microbial structure. The research results can provide a scientific reference for the utilization of SHMS as feed in the goat industry.

饲粮中添加布拉迪酵母对安格斯犊牛生长性能、血清免疫指标及粪便微生物区系的影响

本试验旨在研究饲粮中添加布拉迪酵母对安格斯犊牛生长性能、血清免疫指标及粪便微生物区系的影响。试验选取(152±10)日龄、体重相近的健康安格斯犊牛24头,随机分为3组,每组8头牛。对照组(CG组)饲喂基础饲粮,试验组分别在基础饲粮中添加100(LG组)和200 mg/kg布拉迪酵母(HG组)。预试期14 d,正试期28 d。结果表明:1) LG和HG组犊牛终末体重和平均日增重较CG组差异不显著(P>0.05)。2) LG组犊牛体高较CG组显著提高(P<0.05)。3)LG和HG组犊牛血清白细胞介素-6(IL-6)含量较CG组极显著下降(P<0.01),且HG组显著高于LG组(P<0.05);LG组犊牛血清肿瘤坏死因子-α(TNF-α)含量较CG组显著下降(P<0.05),且HG组极显著高于LG组(P<0.01)。4) LG组Shannon指数较CG组显著上升(P<0.05)。5)在门水平上,3组的优势菌门均为厚壁菌门和拟杆菌门。在科水平上,3组的优势菌科均为瘤胃球菌科、理研菌科、毛螺菌科、普雷沃氏菌科。LG组梭菌目_vadinBB60群的相对丰度显著高于CG组(P<0.05),而丹尼丝菌科的相对丰度显著低于CG组(P<0.05)。HG组丹尼丝菌科的相对丰度显著高于CG组(P<0.05),而梭菌目_vadinBB60群的相对丰度显著低于CG组(P<0.05)。在属水平上,3组的优势菌属均为瘤胃球菌科_UCG-005、理研菌科_RC9_肠道群、瘤胃球菌科_UCG-010、拟杆菌属、艾克曼菌属。LG和HG组dgA-11_gut_group的相对丰度显著低于CG组(P<0.05)。线性判别分析效应量(LEfSe)分析结果显示,LG和HG组芽孢杆菌科、芽孢杆菌属等的相对丰度增加。综上所述,饲粮中添加布拉迪酵母对安格斯犊牛的生长性能无显著影响,可提高犊牛体高,降低血清促炎细胞因子IL-6和TNF-α含量,增强机体免疫功能,提高芽孢杆菌属等有益菌的相对丰度,改善粪便微生物区系,促进犊牛健康。本试验中,饲粮中添加100 mg/kg布拉迪酵母的效果最佳。

为了研究灵芝多糖对断奶川中黑山羊羔羊肝脏免疫功能的影响,试验选取40~50日龄、体重14 kg左右的断奶川中黑山羊公羔48只,随机分为4组,即空白对照(CON)组、低剂量(LGLP)组、中剂量(MGLP)组和高剂量(HGLP)组,每组12只。试验期间分别向CON组、LGLP组、MGLP组和HGLP组基础饲粮中添加0,200,400,800 mg/kg(以干物质计)的灵芝多糖,预试期7 d,正试期56 d。试验结束后,每组随机选择6只川中黑山羊屠宰,分离肝脏用于肝脏免疫指标测定和转录组测序,并对差异表达基因进行GO生物学进程及KEGG代谢通路分析。结果表明:与CON组比,MGLP组(添加400 mg/kg灵芝多糖)可以显著提高川中黑山羊肝脏白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)和IL-1β水平(P<0.05)。MGLP组免疫效果最好,因此选择MGLP组、CON组的肝脏样本进行转录组测序分析。转录组测序分析结果表明,MGLP组与CON组差异表达基因显著富集到抗原加工和提呈、T细胞受体信号通路、趋化因子信号通路和裂原激活蛋白激酶(MAPK)信号通路4条免疫相关通路(P<0.05)。4条免疫相关通路共筛选到19个差异表达基因,CCL5、CⅡTA、LOC102183314等11个差异表达基因显著上调(P<0.05),DUSP1、MYC、LOC102177850等8个差异表达基因显著下调(P<0.05)。说明添加400 mg/kg灵芝多糖可以显著改善断奶川中黑山羊羔羊肝脏免疫功能,并通过调节抗原加工和提呈、T细胞受体信号通路、趋化因子信号通路和MAPK信号通路途径相关基因的表达来调节羔羊肝脏免疫功能。

Abstract :

To further understand the genetic structure that is associated with insulin (INS) and thyroid hormones (TH), including triiodothyronine (T-3) and thyroxine (T-4), in Chinese Holstein cows, we conducted a genome-wide association study (GWAS) of thyroid hormones and insulin in cows. We conducted GWAS analysis on 1217 Chinese Holstein cows raised in southern China and found 19 significant single nucleotide polymorphisms (SNPs) in this study: 10 SNPs were associated with INS, 5 SNPs were associated with T-3, and 4 SNPs were associated with T-4. In our study, the GWAS method was used for preliminary screening on related genes of traits, and due to insufficient relevant literature, a functional analysis of genes could only be based on human studies. We observed thatDGKBfromBos tauruschromosome (BTA)4 is strongly associated with insulin secretion. We found thatEXOC4gene was significantly correlated with T(3)and T(4)traits. Another significant SNP was located in theCYP7A1gene, which has been confirmed to be affected by thyroid hormones.

选取24只体重相近(20 kg±5 kg)的川中黑山羊,随机分为4组,每组6只.对照组育肥羊饲喂不添加碱贮真姬菇菌糠的日粮,试验Ⅰ、Ⅱ、Ⅲ组育肥羊分别饲喂添加20%、30%和40%碱贮真姬菇菌糠的日粮,预饲期14 d,正饲期60 d,研究碱贮真姬菇菌糠对育肥羊生产性能、屠宰性能及血液生化指标的影响.结果表明,在日粮中添加不同水平的碱贮真姬菇菌糠可以降低饲料成本,提高经济效益且对育肥羊的生产性能、屠宰性能及血液生化指标没有不良影响.试验Ⅰ、Ⅱ、Ⅲ组育肥羊的平均日采食量较对照组分别提高了12.28%、16.37%和5.85%(P<0.05);试验组育肥羊的屠宰性能较对照组有提高的趋势,但差异不显著;试验组育肥羊血液中谷胱甘肽过氧化物酶和超氧化物歧化酶的活性较对照组均有提高,但差异不显著;试验Ⅱ组育肥羊的瘤胃重量和瘤胃指数较对照组分别提高了22.00%和12.03%(P<0.05),总胆固醇浓度较对照组提高了28.95%(P<0.05).综合评定结果显示,添加30%碱贮真姬菇菌糠的效果最佳.

Abstract :

We aimed to identify QTL for serum electrolyte traits by performing a GWAS of calcium, chloride, sodium, potassium and magnesium ion concentrations in Chinese Holstein cattle. We detected five SNPs that had significant associations with calcium ion concentrations and identified GATA2 from Bos taurus chromosome (BTA)22 as having the highest significance. Among the genes with significant results, we speculate that TMEM123 might be related to calcium channel proteins according to the functions of the TMEM family.

试验旨在研究碱贮对真姬菇菌糠瘤胃降解率的影响,采用CaO、Ca(OH)_2、CaO+3%尿素及Ca(OH)_2+3%尿素4种处理,每种处理碱(CaO/Ca(OH)_2)的添加量设3%、4%、5%及6%4个水平,分别记作CO3、CO4、CO5、CO6,CH3、CH4、CH5、CH6,COU3、COU4、COU5、COU6,CHU3、CHU4、CHU5、CHU6,每个水平4个重复,空白组设2个,1组空白(CK)仅为真姬菇菌糠,另1组空白(CKU)为真姬菇菌糠+3%尿素,室温下碱贮30 d。选取6只装有瘤胃永久瘘管川中黑山羊作为瘤胃液供体,对真姬菇菌糠的干物质(DM)、中性洗涤纤维(NDF)、酸性洗涤纤维(ADF)、粗蛋白质(CP)体外发酵4、8、12、24、48、72 h的动态降解率进行测定与分析。结果表明,真姬菇菌糠经不同水平碱贮后,DM、CP、NDF、ADF的72 h降解率及其有效降解率(ED)均较对照组提高。其中,COU碱贮处理组效果最佳,不同水平COU碱贮处理组的72 h的DM瘤胃降解率较CKU组分别提高13.01%、20.03%、31.65%和32.33%(P<0.05);NDF瘤胃降解率较CKU组分别提高27.61%、45.78%、62.88%和50.23%(P<0.05);ADF瘤胃降解率较CKU组分别提高26.14%、45.75%、51.76%和39.65%(P<0.05);CP瘤胃降解率较CKU组分别提高12.33%、14.62%、20.55%和19.07%(P<0.05),其中综合评定COU5处理水平最优。
