Schisandra chinensisbee pollen's chemical profiles and protective effect against H2O2-induced apoptosis in H9c2 cardiomyocytes SCIE

Abstract :

Background Schisandra chinensis(Turcz.) Baill bee pollen extract (SCBPE) is often used as a functional food in China due to its good antioxidant property. However, its chemical compositions and effects on H9c2 cardiomyocytes against H2O2-induced cell injury still lacks of reports thus far. This study aimed to characterize the main components of SCBPE and investigate its protective effects against H2O2-induced H9c2 cardiomyocyte injury. Methods The main components of SCBPE were analyzed via ultraperformance liquid chromatography-quadrupole time-of-flight tandem mass spectrometry (UPLC-QTOF MS/MS). The three main nucleosides in SCBPE were quantitatively analyzed via ultraperformance liquid chromatography-diode array detection. Furthermore, the potential mechanism by which SCBPE exerts protective effects against H2O2-induced H9c2 cardiomyocyte injury was explored for the first time via cell survival rate measurements; cell morphological observation; myocardial superoxide dismutase (SOD) activity and malondialdehyde (MDA) and glutathione (GSH) level determination; flow cytometry; and quantitative polymerase chain reaction. Results Two carbohydrates, three nucleosides, and nine quinic acid nitrogen-containing derivatives in SCBPE were identified or tentatively characterized via UPLC-QTOF MS/MS. The nine quinic acid nitrogen-containing derivatives were first reported in bee pollen. The contents of uridine, guanosine, and adenosine were 2.4945 +/- 0.0185, 0.1896 +/- 0.0049, and 1.8418 +/- 0.0157 mu g/mg, respectively. Results of in vitro experiments showed that cell survival rate, myocardial SOD activity, and GSH level significantly increased and myocardial MDA level significantly decreased in SCBPE groups compared with those in H(2)O(2)group. Cell morphology in SCBPE groups also markedly improved compared with that in H(2)O(2)group. Results indicated that SCBPE protected H9c2 cardiomyocytes from H2O2-induced apoptosis by downregulating the mRNA expressions of Bax, cytochrome C, and caspase-3 and upregulating the Bcl-2 mRNA expression. Conclusions This study is the first to report that SCBPE could protect against oxidative stress injury and apoptosis in H2O2-injured H9c2 cells. Results indicated that the nucleosides and quinic acid nitrogen-containing derivatives could be the main substances that exert protective effects against H2O2-induced H9c2 cardiomyocyte injury.

拟设计在体内低毒、靶向肿瘤细胞的抗癌多肽(记:GPG),克服在抗癌治疗中使用化学类药物存在的缺点,探讨用荧光光谱评价靶向肽对肿瘤肿块包覆状况的方法;进行仅用一组基于双报告基团的小鼠模型(用EGFP转染的鼠肝肿瘤细胞H_(22),记:H_(22)-EGFP与荧光染料Cy7标记的GPG,记:Cy7-GPG,作报告基团),就能完成抗癌肽体内主要性能监测的研究。用H_(22)-EGFP构建小鼠移植瘤模型,尾静脉注射Cy7-GPG后,用成像仪(Ex=750 nm)观察到肿瘤的荧光光子数不断增大,从第4 h的(3.90±0.260)×10~6 photons·(s·cm~2)~(-1)(单位下同)升至第24 h的(1.28±0.330)×10~8,橙色荧光全部聚集到肿瘤上,而对照组中Cy7的荧光没有聚集在肿瘤上,且肿瘤上光子数也无明显变化;此时,用成像仪在实验组同一只鼠上,分别用750和488 nm激发波长录得包覆状物和实际肿瘤肿块的图片,表明它们的大小和形状一致;以上两项测定后,再每2 d一次往上述实验组鼠体内注射GPG,在成像仪上(Ex=488 nm)监测到它们的肿瘤变小,荧光光子数逐渐降低,从第2 d的(4.15±0.291)×10~6降至第56 d的(4.75±0.283)×10~4,空白对照组正相反。GPG与化学药物环磷酰胺抗癌活性相当,但后者对小鼠毒副作用严重;最后,往上述GPG药效实验组鼠体内注射Cy7-GPG, 48 h后处死小鼠,取出内脏器官及肿瘤肿块,在成像仪上(Ex=750 nm)录得它们的荧光光子数。实验表明GPG靶向性强、对肿瘤肿块包覆程度高、药效强,对其他主要脏器无毒副作用。本文构建的基于双报告基团荧光成像模型,可监测靶标被包覆状况,克服了传统只能对靶向肽靶向功能进行评价的弊端,增进对药物作用机理的认识;本文仅消耗一组实验鼠模型,节约实验成本、简捷地监测了靶向肽主要性能,表明GPG在体内性能较好,具有应用价值。

【目的】探讨在巯乙基磺酸钠(MESNa)与碳酸氢铵(NH_4HCO_3)存在下,对在大肠杆菌上表达的重组融合蛋白中内含肽(Intein)用硫醇(DTT)进行自剪切,促使抗菌肽(MME)碳末端实现酰胺化。【方法】构建用大肠杆菌表达内含肽介导的MME重组质粒,通过表达获得依次由"组氨酸、Sumo标签、MME、内含肽"构成的融合蛋白,用镍柱及透析进行纯化,在MESNa和NH_4HCO_3存在下,用DTT对内含肽进行自剪切,促使MME碳末端酰胺化,肠激酶切割、纯化,得到碳末端酰胺化的MME。质谱和二级串联质谱配合使用,检测MME及其碳末端酰胺化。【结果】通过PCR,鉴定融合蛋白,其基因片段为837 bp,符合预期。质谱鉴定得MME相对分子量为3 057.7与其理论值3 057.64一致。二级串联质谱检测得MME碳端碎片的分子量为1 214.728,与碳末端酰胺化的MME碳端碎片理论分子量1 214.739相符,匹配度为45,表明本研究制备的MME碳末端已被酰胺化,该蛋白为可溶。【结论】用大肠杆菌成功地表达了重组融合蛋白,在MESNa与NH_4HCO_3存在下,促进了内含肽自剪切,MME碳末端被酰胺化,实现了简单的"一步法"制备。质谱与二级串联质谱配合使用,可灵敏、简单地对碳末端酰胺化多肽进行检测,可作为该项检测方法的一种选择,结果表明,本研究成功地制备了碳末端酰胺化的MME。

为获得低毒、具有抗肿瘤和抗菌活性的长效改性蜂毒肽,提高蜂毒肽的临床应用价值,本试验以多肽构效关系为指导,选择两亲性α-螺旋型阳离子多肽分子结构为改性蜂毒肽的设计模板,用杂合肽法设计了一系列多肽序列,用生物信息学工具比较它们的生物活性并进行初选,再用固相法合成初选所得少量序列的多肽,比对合成多肽的生物活性,精选获得生物活性高的多肽。实验表明精选所得多肽具有长效性,确定其为设计目标肽(GPG),当GPG浓度为150μmol/L时,其溶血率为零;浓度为33.3μmol/L时,其对小鼠肝癌细胞(H_(22))、人肝癌细胞(SMMC-7721)和人结肠癌细胞(SW-1116)的抑癌率分别为(78.0±0.6)%、(74.0±1.2)%、(74.0±0.5)%;浓度为100μmol/L时,其对大肠杆菌的抑菌圈直径为(15.18±0.30) mm;当注射GPG(100μmol/只)时,能延长致死剂量大肠杆菌(5×107个/只)攻毒的小鼠寿命63.2 h。该设计省时省力地优选得溶血活性低、抗癌抗菌活性强、半衰期长的全新结构目标肽GPG,其有临床应用前景;以GRGDSP肽链为多肽氮端端基,可提高其生物活性及半衰期。

Abstract :

Oxidative stress plays an important role in the pathogenesis of myocardial infarction (MI). Schisandra chinensis bee pollen extract (SCBPE) possesses powerful antioxidant capacity. This study aimed to further explore the antioxidative and cardioprotective effects of SCBPE on acute MI induced by isoprenaline (ISO) in rats. The rats were intragastrically administrated with SCBPE (600, 1200, or 1800 mg/kg/day) and Compound Danshen dropping pills (270 mg/kg/day) for 30 days, then subcutaneously injected with ISO (65 mg/kg/day) on the 29th and 30th day. Compared with the model group, pretreatment with middle and high doses of SCBPE significantly reduced serum aspartate transaminase, lactate dehydrogenase, and creatine kinase activities and increased myocardial superoxide dismutase, glutathione peroxidase, and catalase activities. The histopathologic aspects showed that pathological heart change was found in the model group and reduced to varying degrees in the SCBPE groups. Moreover, the protein expression of nuclear factor-erythroid 2-related factor 2 (Nrf-2), heme oxygenase-1 (HO-1), and Bcl2 in the heart increased in the SCBPE groups, while that of Bax decreased compared to the model group. Besides this, uridine was isolated from S. chinensis bee pollen for the first time. This study could provide a scientific basis for using Schisandra chinensis bee pollen as a functional food for the prevention of MI.

Apis cerana is an endemic species of honey bee in Asia. A. cerana is believed to have better adaptation to scattered nectar sources than Apis mellifera, which implies that honey can be harvested from A. cerana colonies, but A. mellifera will starve at the same location. We hypothesize that there are differences in foraging performance, worker longevity, and daily sugar consumption between A. cerana and A. mellifera workers. These parameters in both species were determined. A. cerana had longer longevity than A. mellifera if longevity was measured in caged bees without pollen or in mixed species colonies with A. mellifera queens, but A. mellifera had much longer longevity when measured inside cages provided with pollen. A. cerana also had more foraging trips and higher proportion of foragers than A. mellifera in mixed colonies. The daily sugar consumption of A. cerana workers was 32% of that of A. mellifera, perhaps mainly due to the smaller biomass of A. cerana workers. In addition, A. cerana workers showed significantly lower variance in daily sugar consumption. In conclusion, A. cerana has more foraging trips, higher forager proportion, less sugar consumption, and longer survival than those of A. mellifera, when measured inside the same colonies. These differences might explain why during a dearth or in scattered resource areas, A. cerana can store or produce more honey than A. mellifera.

蜂胶与β-环糊精复合物在西番莲果汁饮料中的应用研究 PKU

为改善蜂胶的水溶性,将其添加到西番莲果汁饮料中,研制一款功能性西番莲果汁饮料。将β-环糊精(β-cyclodextrin,β-CD)与蜂胶乙醇提取物(ethanol extract of propolis,EEP)制成复合物(compound of ethanol extract of propolis and beta cyclodextrin,EEP-βCD),测定EEP-βCD的水溶性和热稳定性,并对西番莲果汁饮料的工艺参数进行优化。结果表明:EEP与β-CD复合后,水溶性和热稳定性都显著提高(P <0.01);西番莲果汁饮料的最佳工艺参数为:西番莲果汁15%,木糖醇12%,EEP-βCD 0.6%。蜂胶与β-环糊精复合后水溶性、热稳定性和刺激性味道都得到改善,可将其应用于西番莲果汁饮料。

为研究从蜂蜜中分离的鲁氏接合酵母在高糖培养基中的生理特性变化,本文对鸭脚木蜜的耐高渗酵母菌进行平板分离,筛选具有代表性的酵母菌菌落,摇培后提取其DNA,进行ITS序列扩增,经比对后获得酵母菌的种属信息;用30%、40%、50%、60%的高糖培养基进行培养,测定其生长、培养基残糖量和氧化酶活力的变化。分离鉴定后得到鲁氏接合酵母,与对照组相比,在40%、50%、60%组中生长速率降低,但30%培养基中出现二次生长;随着糖浓度的增加,酵母菌的糖耗量减少,在30%培养基中糖耗量最大;但在高糖培养基中酵母菌胞内SOD、POD、CAT活力增加。这些结果为筛选耐糖工程菌提供理论依据。
