Abstract :
High light (HL) significantly impacts plant photosynthesis. This study investigated the effects of different magnesium (Mg) levels (0, 1, 2, and 5 mol Mg plant-1; HMg0, HMg1, HMg2, and HMg5) on tobacco (Nicotiana tabacum L. cv. Cuibi No. 1) under HL (1500 mu mol m-2 s-1), aiming to understand the role of Mg in mitigating the impact of HL on photosynthesis and carbon-nitrogen metabolism. Plants treated with 1 mol Mg plant-1 under 750 mu mol m-2 s-1 light conditions served as the control. HL led to a reduced chlorophyll (Chl) content and inhibited the maximum photosynthetic rate (Pmax). It also decreased energy involved in photosynthetic electron transfer (ET) and electron flux to reduction end-electron acceptors at the photosystems I (PSI) acceptor side (RE) and caused photosynthetic system damage. H2O2 accumulation exacerbated membrane lipid peroxidation damage, disrupting carbon and nitrogen metabolism, and inducing antioxidant enzyme activity. HMg2 increased Chl content, stomatal conductance, intercellular CO2 concentration, and the net photosynthetic rate compared to HMg0. It enhanced ET efficiency, PSI and PSII functionality, reduced dissipated energy flux (DI), and minimized photosynthesis damage. Conversely, excessive Mg application (HMg5) decreased Pmax and PSII activity, increasing DI. Adequate Mg supply alleviated HL's detrimental effects by enhancing Chl content and ET and RE efficiency.

BackgroundBoron (B) is an essential micronutrient for plants. Inappropriate B supply detrimentally affects the productivity of numerous crops. Understanding of the molecular responses of plants to different B supply levels would be of significance in crop improvement and cultivation practices to deal with the problem.ResultsWe conducted a comprehensive analysis of the transcriptome and proteome of tobacco seedlings to investigate the expression changes of genes/proteins in response to different B supply levels, with a particular focus on B deficiency. The global gene and protein expression profiles revealed the potential mechanisms involved in the responses of tobacco to B deficiency, including up-regulation of the NIP5;1-BORs module, complex regulation of genes/proteins related to cell wall metabolism, and up-regulation of the antioxidant machinery.ConclusionOur results demonstrated that B deficiency caused severe morphological and physiological disorders in tobacco seedlings, and revealed dynamic expression changes of tobacco genes/proteins in response to different B supply levels, especially to B deficiency, thus offering valuable insights into the molecular responses of tobacco to B deficiency.

Introduction: Magnesium (Mg) is a crucial macronutrient for plants. Understanding the molecular responses of plants to different levels of Mg supply is important for improving cultivation practices and breeding new varieties with efficient Mg utilization. Methods: In this study, we conducted a comprehensive transcriptome analysis on tobacco (Nicotiana tabacum L.) seedling leaves to investigate changes in gene expression in response to different levels of Mg supply, including Mg-deficient, 1/4-normal Mg, normal Mg, and 4x-normal Mg, with a particular focus on Mg deficiency at 5, 15 and 25 days after treatment (DAT), respectively. Results: A total of 11,267 differentially expressed genes (DEGs) were identified in the Mg-deficient, 1/4-normal Mg, and/or 4x-normal Mg seedlings compared to the normal Mg seedlings. The global gene expression profiles revealed potential mechanisms involved in the response to Mg deficiency in tobacco leaves, including down-regulation of genes-two DEGs encoding mitochondria-localized NtMGT7 and NtMGT9 homologs, and one DEG encoding a tonoplast-localized NtMHX1 homolog-associated with Mg trafficking from the cytosol to mitochondria and vacuoles, decreased expression of genes linked to photosynthesis and carbon fixation at later stages, and up-regulation of genes related to antioxidant defenses, such as NtPODs, NtPrxs, and NtGSTs. Discussion: Our findings provide new insights into the molecular mechanisms underlying how tobacco responds to Mg deficiency.

为了完善烟稻轮作烟区养分管理和促进烤烟生长后期烟叶“养熟”措施优化,通过大田试验,在水稻季和烤烟季设置T1(烤烟季水稻季都施用纯化肥)、T2(烤烟季化肥+饼肥,水稻季纯化肥)、T3(烤烟季纯化肥,水稻季化肥+饼肥)、T4(烤烟季水稻季都施用化肥+饼肥)4种施肥处理,研究烟稻轮作烟田不同作物季施用有机肥对烟株生长后期光合特性、荧光特性以及碳氮代谢关键酶活性的影响。结果表明,施用饼肥处理各项指标均比2季都施用纯化肥处理好,饼肥在烤烟当季施用比水稻季施用对烟株生理效应更好,烟稻2季均施用化肥+饼肥处理对烟株生理效应最好,T4处理总叶绿素含量较T2、T3、T1处理分别增加15.0%、26.6%、45.3%,T4处理净光合速率较T2、T3、T1处理分别增加13.3%、36.1%、66.0%。烟稻2季均化肥饼肥配施能更显著的提高光合速率,促进光合产物的转化以及维持氮代谢水平,保证烤烟生长后期烟株体内各种代谢过程的顺利进行,更有利于烟叶在田间“养熟”和优质烟叶的品质形成,从而提高烟农收益。

为了解不同形态氮素与不同镁浓度对烤烟早期光合荧光特性及生长的影响,以翠碧一号烤烟为材料,采用水培试验方法,在温度较低、光照较弱条件下设置3种氮素处理和两个镁水平,分析不同处理烟株光合荧光特性及生长的差异。结果表明,加镁条件下铵、硝态氮各50%处理烟株的生物量、最大净光合速率(P_(max))、表观量子效率(AQY)和光系统Ⅱ(PSⅡ)与光系统Ⅰ(PSⅠ)及系统间电子传递链的整体功能活性指数(PI _(total))均最高,其中生物量相较于纯硝态氮且加镁处理提高21.7%,相较于纯铵态氮加镁处理提高96.0%;不加镁条件下全硝态氮处理P_(max)和AQY高于铵、硝态氮各50%处理且显著高于全铵态氮处理。纯硝态氮处理和铵、硝态氮各50%处理加镁后显著提高各项指标,但加镁对全氨态氮处理没有显著性影响。可见,在温度较低、光照较弱条件下,铵、硝各50%且加镁处理更有利于烤烟早期的生长发育。

Zinc (Zn) is an essential micronutrient for plants. Adequate regulation of Zn uptake, transport and distribution, and adaptation to Zn-deficiency stress or Zn-excess toxicity are crucial for plant growth and development. However, little has been done to understand the molecular responses of plants toward different Zn supply levels. In the present study, we investigated the growth and physiological responses of tobacco seedlings grown under Zn-completely deficient, Zn-limiting, Zn-normal, and Zn-4-fold sufficient conditions, respectively, and demonstrated that Zn deficiency/limitation caused oxidative stress and impaired growth of tobacco plants. Combined transcriptome and proteome analysis revealed up-regulation of genes/proteins associated with Zn uptake and distribution, including ZIPs, NAS3s, and HMA1s, and up-regulation of genes/proteins involved in regulation of oxidative stress, including SODs, APX1s, GPX6, and GSTs in tobacco seedlings in response to Zn deficiency/ limitation, suggesting that tobacco possessed mechanisms to regulate Zn homeostasis primarily through upregulation of the ZIPs-NAS3s module, and to alleviate Zn deficiency/limitation-induced oxidative stress through activation of the antioxidant machinery. Our results provide novel insights into the adaptive mechanisms of tobacco in response to different Zn supplies, and would lay a theoretical foundation for development of varieties of tobacco or its relatives with high tolerance to Zn-deficiency.

为明确福建‘翠碧一号’烤烟适宜的施镁量以及施镁对烟田土壤供镁的影响,采用田间试验研究不同施镁量对烟田不同时期土壤供镁、烟株物质积累和烤后烟叶产质量的影响。结果表明,随施镁量的增加,土壤交换性镁浓度上升,不同时期土壤供镁能力与施镁量表现为显著的直线线性相关关系。终采时烟株不同部位叶片镁含量和积累量也均与施镁量存在显著的直线线性相关关系。施镁处理的均价、产值、上等烟比例和上中等烟比例分别比不施镁处理提高了6.39%~11.67%、6.13%~14.63%、18.43%~31.46%和5.36%~8.96%。随施镁量的增加烤后烟叶总钾含量呈下降趋势。综合烤后不同部位烟叶感官评吸质量以施镁(以MgO计)量在19.5 kg/hm~2处理最好,其次为39 kg/hm~2处理;产值以施镁58.5 kg/hm~2处理最高。施镁量对烟株生物量积累和烤后烟叶产量的影响规律性不明显。综合福建植烟土壤交换性镁含量的实际情况,以及土壤供镁、烟株物质积累和烤后烟叶质量,认为福建‘翠碧一号’烤烟生产中,施镁(以MgO计)量以19.5~39 kg/hm~2为宜,既能保证土壤正常供镁和镁营养的相对平衡,又能满足烟株对镁营养的需求,提高烤后烟叶质量。

为研究烤烟生产过程中适宜的钾镁比例,通过开展田间随机区组试验,以农户常规施肥(N∶P_2O_5∶K_2O=1∶0.68∶3,施K_2O量为315 kg/hm~2)为对照,研究了优化钾肥处理(N∶P_2O_5∶K_2O=1∶0.68∶2,施K_2O量为210 kg/hm~2),优化钾肥+MgO 19.5 kg/hm~2处理,以及优化钾肥+MgO 39.0 kg/hm~2处理对烟株成熟期光合特性、荧光特性和碳氮代谢酶活性的影响。结果表明:降低钾镁比例,烤烟各处理叶绿素中Chl含量的增幅分别为2.2%、11.8%、18.3%;Car含量的增幅分别为3.5%、17.5%、19.7%;最大荧光(F_m)的增幅分别为13.3%、18.0%、24.1%;可变荧光(F_v)的增幅分别为17.3%、23.6%、30.9%;F_v/F_o的增幅分别为11.0%、17.2%、23.1%;F_v/F_m的增幅分别为3.7%、4.7%、5.5%。活性反应中心效率ET_o/ABS、ET_o/TR_o上升,单位面积电子传递的有效性(ET_o/CS_m)提高,减少荧光产生(TR_o/RC、ABS/RC下降),提升采收期烤烟光反应系统功能的协调性,提高光反应系统的整体活性(PI_(ABS)、PI_(total)上升)。RuBP羧化酶(RuBPCase)、谷氨酰胺合成酶(GS)、硝酸还原酶(NR)、蔗糖合成酶(SS)的活性均以优化钾肥+MgO 19.5 kg/hm~2处理最高。优化钾镁比例可以提高光反应系统的活性,提高烤烟的光合作用效率,对平衡碳氮代谢具有积极作用,以钾镁比例为10.77∶1左右、优化钾肥+MgO 19.5 kg/hm~2处理下生理代谢最佳。
