龙眼果实采后果皮褐变过程差异表达蛋白分析 CSCD PKU

Abstract :

【目的】探讨龙眼果实采后果皮常温褐变过程的差异表达蛋白变化。【方法】以‘乌龙岭’龙眼为研究对象,龙眼果实采后在常温(28±2)°C下贮藏0、1、3和5 d后,应用双向电泳和MALDI-TOF/TOF-MS技术分析果皮的差异蛋白。【结果】在龙眼常温下贮藏0、1、3和5 d等不同时间点,共检测到42个差异蛋白至少发生一次丰度在2倍以上的表达变化,其中38个蛋白被成功鉴定,有14个上调表达,20个下调表达,4个复杂变化。这些蛋白质参与了应激和防御(13.2%)、蛋白折叠、稳定和降解(28.9%)、能量与物质代谢(26.3%)、信号转导(10.5%)和次生代谢(2.6%)等生理过程。【结论】脱氢抗坏血酸还原酶、过氧化氢酶、蛋白质二硫键异构酶的下调表达,可能引起龙眼果实采后果皮细胞内活性氧增加;苹果酸酶和膜联蛋白的下调表达,可能与生物膜系统完整性的破坏有关;另外泛素结合酶、蛋白酶体α亚基和20S蛋白酶体PAF1亚基的下调表达,可能导致对果皮细胞中损伤或失活蛋白的选择性降解速率降低,使细胞内环境平衡失调;而过氧化物酶的上调表达,可能引起酚类物质反应生成褐色物质,致使果皮发生褐变。

Abstract :

A comparative proteomic approach was carried out to investigate anthocyanin biosynthesis in the tuberous roots of yellow sweet potato (YSP) and purple sweet potato (PSP) cultivars. More than 800 proteins were reproducibly detected through two-dimensional electrophoresis (2-DE), of which 50 proteins with 39 more and 11 less accumulated in PSP were identified through matrix-assisted laser desorption ionization-time of flight/time of flight mass spectrometry (MALDI-TOF/TOF-MS). Most of the analyzed proteins are annotated to be involved in starch metabolism and glycolysis. The more abundant starch phosphorylase (SP) and phosphoglucomutase (PGM) in PSP promoted the synthesis of precursors for anthocyanin synthesis. The results implied that starch degradation provided abundant substrates for anthocyanin biosynthesis in tuberous roots of PSP. 24 kDa vacuolar protein (VP24) is related to anthocyanin transport and accumulation in vacuoles. Vacuole-associated annexin protein, VCaB42, is correlated with tonoplast biogenesis. Synergistic action of the two proteins is probably involved in the microautophagy and the intravacuolar trapping of anthocyanins. Interestingly, both VCaB42 and VP24 were more accumulated in PSP, suggesting that anthocyanins generated in the cytosol were transported into and became stored in the vacuoles of PSP. The present study provides new insights into the mechanism of tuberous root-specific anthocyanin accumulation in PSP. Biological significance: Sweet potato ranks as the seventh most important crop worldwide. Purple sweet potato, a special sweet potato cultivar, has been extensively investigated because large amounts of anthocyanin accumulate in its tuberous roots. Anthocyanin is well known for its free radical-scavenging activity and beneficial effects on human health. Its biosynthetic pathway has been well characterized in model plants. Although large-scale systematic studies have been performed to identify the proteins present in sweet potato, information on the regulation of anthocyanin synthesis in sweet potato is insufficient. Our proteome study demonstrated that starch degradation may contribute to anthocyanin accumulation in purple sweet potato. To our knowledge, this study is the first to propose that starch degradation may provide precursors of anthocyanin biosynthesis in sweet potato. (C) 2016 Elsevier B.V. All rights reserved.

Abstract :

This study developed a proteome reference map of Myrica rubra fruits at the green, pink and red stages during ripening using two-dimensional gel electrophoresis (2-DE). Forty-six differentially expressed proteins were detected in the gel, of which 43 were successfully identified by matrix-assisted laser desorption ionization time-of-flight/time-of-flight mass spectrometry and protein database searching. We found that malic enzyme related to the decrease of organic acid acidity was up-regulated. The high abundance of pyruvate decarboxylase and alcohol dehydrogenase may contribute to fruit peculiar fragrant characteristics. Phenylalanine ammonia-Iyase, chalcone synthase 11, UDP-glucose:flavonoid 3-O-glucosyltransferase, and anthocyanidin synthase, enzymes involved in the anthocyanin metabolic pathway, were all up-regulated. The physiological data agree with fruit proteome results. These findings provided insights into the metabolic processes and regulatory mechanisms during Chinese bayberry fruit ripening. Abstract :

本文通过双向电泳技术结合质谱分析的方法研究常温贮藏采后3 d的龙眼果皮蛋白,分离出800多个可重复蛋白,成功鉴定7个差异表达2倍以上并与氧化胁迫相关的差异蛋白,它们大都下调表达。其中,脱氢抗坏血酸还原酶和过氧化氢酶显著下调表达,表明龙眼果实采后3 d清除活性氧(ROS)的能力显著下降,导致细胞膜系统的完整性被破坏。同时,引起褐变的主要酶之一的过氧化物酶明显上调表达。这些结果表示,龙眼果实采后保持一定的抗氧化和清除ROS能力是延缓果皮褐变的重要措施。

Abstract :

为探索龙眼(Dimocarpus longan)果肉常温劣变过程中蛋白质的变化,采用蛋白质组学的方法,在‘福眼’龙眼果肉常温劣变过程中发现共有24个2倍差异表达蛋白,其中21个蛋白被成功鉴定。这些蛋白参与了应激反应与防御(56%)、能量与碳代谢(19%)、初级代谢(5%)、蛋白转运(5%)和细胞骨架(5%)等细胞代谢活动。大多数与抗氧化作用密切相关的蛋白质都下调表达,说明龙眼采后果肉的抗氧化能力有所下降,不能有效地缓解ROS积累引起的毒性作用,从而使果肉劣变。此外,与能量代谢相关的蛋白全部下调表达,表明龙眼果实采后果肉劣变可能与能量供应不足有关。这些结果为龙眼采后保鲜技术研究提供科学依据。

以‘乌龙岭’龙眼品种为研究对象,分析了龙眼干制过程中糖类含量的动态变化。结果表明,干制过程中龙眼果肉褐变度显著增加,含水量明显下降,果糖、葡萄糖和蔗糖在干制初期均显著性下降,说明干制过程果肉的褐变与糖类含量、含水量变化关系密切;在干制过程中果肉可溶性总糖和还原糖含量均显著下降,分别下降了36.38%、42.09%,这主要由于单糖和寡糖的大量消耗形成深色物质;贮藏1年的龙眼干果实中碳水化合物下降,因此,食用龙眼干时要注意保质期。

Abstract :

为探明烟草青枯菌在不同杂草根部的寄生和腐生状况以及冬季在杂草根部的消长和传病能力,采用抗利福平青枯菌接种烟草诱发青枯病,将不同的带菌杂草根埋入无病盆土并移栽烟草监测青枯病发生,同时以含利福平的选择性培养基对49种杂草根、杂草的不同部位、病土中杂草烂根和冬季病土中杂草根作带菌测定。腋花蓼、香附子等7种杂草根感青枯菌;无芒稗、龙葵等18种根为中感;黄鹌菜、稗草等24种根为低感。鬼针草和商陆仅根表带菌;早熟禾和无芒稗等6种根内外带菌;牛筋草和马齿苋等3种根、茎、叶带菌。青枯菌能在牛筋草和水蓼等8种杂草的烂根中高度腐生,在无芒稗和座地菊等7种烂根中中度腐生,在石龙芮和稀签等6种烂根中低度腐生。红辣蓼等7种杂草均能诱发烟草青枯病,其中红辣蓼、空心莲子草等4种杂草带菌的根传病能力强,发病率85.7%~100%。烟草青枯菌能在杂草根部定殖,根腐烂时能继续生长繁殖,并且能在早熟禾等6种杂草根部安全越冬,成为翌年有效的初侵染源,引发烟草青枯病,其中看麦娘处理的发病早且重,这是导致烟草青枯病发生与流行的一个重要因素。

Colonization and infection of Ralstonia Solanacearum in weed roots EI CSCD PKU

Abstract :

Parasitism and saprophytism of Ralstonia Solanacearum in different weeds were studied to provide basis for preventing tobacco bacterial wilt. Growth and decline trends and infectivity ability of the bacteria in winter were monitored. Disease was induced by inoculating tobacco seedlings with rifampicin-resistant R. solanacearum. Bacteria in 49 weed roots in tobacco planting land. Weed roots from infected soil in winter were determined using a selective culture medium containing rifampicin. The occurrence of tobacco bacterial wilt was monitored by planting tobacco seedlings in pots with uninfected soil. 7 weeds such as Polygonum plebeium R.Br and Cyperus rotundus L. were susceptible, 18 such as Pospalum orbiculare Forst. and Solanum mgrum L. were moderately susceptible, while 24 weeds such as Youngia japonica (Linn.) DC and Echinochloa crusgalli (L.) Beauv. were low susceptible. R.Solanacearum were just noted on the root surface of Bidens bipinnata L. and Phytolacca acinosa Raxb., but were both noted in vivo and in vitro of 6 weed roots such as Poa annua L. and Pospalum orbiculare Forst..3 weeds, i.e. Eleusine indica (L.) Gaertn and Portutulaca oleracea. R.Solanacearum was highly saprophytic in 8 rotten roots such as Eleusine indica (L.) Gaertn and Polygonum hydropiper L., moderately saprophytic in 7 rotten roots such as Pospalum orbiculare Forst. and Soliva anthemifolia (Juss), low saprophytic in 6 rotten roots such as Ranunculus sceleratus L. and Siegesbeckia orientalis L. Result showed R.Solanacearum can overwinter in roots of 6 annual weeds and then cause tobacco bacterial wilt in the following year. The disease occurrence can be induced by 7 weeds, while the disease was early and serious in Alopecurus aequalis Sobol. 4 weeds such as Polygonum orientale L. and Altemanthera philoxeroides (Mart) Griseb had the strongest infectivity ability, the morbidity was 85.7%~100%. R. Solanacearum can colonize in weed roots, then proliferate during root decay and overwinter in the weed roots such as Alopecurus aequalis Sobol, and finally become the effective resources of primary infection. It was an important factor that caused the occurrence and epidemic of tobacco bacterial wilt.

龙眼成花逆转花芽α-tubulin基因的克隆与原核表达 CSCD PKU

运用蛋白质组学方法比较龙眼(Dimocarpus longanLour.)正常成花和成花逆转花芽的差异蛋白质组,并应用RACE方法克隆其中上调表达的α-微管蛋白基因α-tubulin,获得一段长度为1641 bp的cDNA,其中包括1个1350 bp的开放阅读框[GenBank登录号:FJ479617(GI:218202929)]。将α-tubulin全长cDNA在大肠杆菌中表达,获得1个约49.6 kD的外源蛋白,经Western blotting验证为α-微管蛋白。RT-PCR和Western blotting分别检测了α-tubulin在转录和翻译水平上的表达,结果表明,α-微管蛋白在成花逆转的龙眼花芽中上调表达,可能是逆转花芽形态差异表现的原因之一。
