姜黄素对过氧化氢诱导的罗非鱼肝细胞热休克蛋白基因表达的影响

Abstract :

【目的】探究热休克蛋白基因在氧化应激和姜黄素作用下的表达差异,了解姜黄素药效的发挥与热休克蛋白基因表达变化的关系,进一步探讨热休克蛋白对氧化应激条件下鱼类肝细胞中的功能,为鱼类氧化应激的毒理学机制及姜黄素的药理学机制提供理论资料。【方法】对24 h姜黄素预处理(保护组)与无姜黄素预处理(对照组)的罗非鱼肝细胞分别进行0、1、2.5 h的H_2O_2应激,并对肝细胞5种高分子热休克蛋白基因(HSP70、HSP90α、HSP90β、HSP60、HSP75)和5种小分子热休克蛋白基因(HSP30、HSPB1、 HSPB7、HSPB8、HSPB11)的表达水平进行qPCR检测。【结果】与不经H_2O_2处理(0 h)相比,H_2O_2作用1 h能显著提升HSPB1表达量,降低HSP30表达量;H_2O_2作用2.5 h能显著提升HSP90a、 HSPB1的表达量。姜黄素预处理24 h能显著提升HSP70、 HSP90α、HSP30、HSPB1的表达量;姜黄素预处理24 h后再进行H_2O_2应激1 h时,HSP70、HSP90α、HSP30的表达量显著高于H_2O_2单独应激(对照组);姜黄素预处理24 h后再进行H_2O_2应激2.5 h时,HSP90α、HSP30、HSPB1的表达量显著高于H_2O_2单独应激。其余热休克蛋白在试验过程中无显著差异。【结论】H_2O_2单独应激可引起罗非鱼肝细胞HSP90a、HSP30、HSPB1基因的表达量变化,而姜黄素预处理能提高多种热休克蛋白基因的表达,提高了罗非鱼肝细胞的抗氧化能力,最终抵御H_2O_2引起的氧化应激并维持细胞活力。

Abstract :

为了研究在饲料中添加两种植物提取物复合物对断奶仔猪血常规和血液生化、血清激素指标的影响,试验选取胎次相近(3~6胎次)、25日龄的断奶“Y×L”二元仔猪120头,按体重相近原则随机分为对照组(A组)、抗生素组(B组)、植物提取物复合物Ⅰ组(C组)和植物提取物复合物Ⅱ组(D组),每组3个重复,每个重复10头。A组饲喂基础日粮,B组在基础日粮中添加20 mg/kg硫酸黏杆菌素+100 mg/kg杆菌肽锌,C组、D组分别在基础日粮中添加400 mg/kg植物提取物复合物Ⅰ、400 mg/kg植物提取物复合物Ⅱ,试验期30 d,分别于试验第15,30天采集试验猪血液,检测血常规和血液生化指标、激素指标。结果表明:第30天,与A组相比,B组、D组红细胞数(RBC)显著或极显著升高(P<0.05或P<0.01)。第15天,与A组相比,B组、D组尿素氮(BUN)含量显著降低(P<0.05);C组总蛋白(TP)含量显著升高(P<0.05);B组、C组、D组球蛋白(GLB)含量显著或极显著降低(P<0.05或P<0.01)。第30天,与A组相比,D组三酰甘油(TG)、总胆固醇(TCHO)、BUN含量均显著降低(P<0.05);C组白蛋白(ALB)含量显著升高(P<0.05)。试验第15,30天,与A组相比,B组、C组、D组血清生长激素(GH)和胰岛素样生长因子-Ⅰ(IGF-Ⅰ)含量显著或极显著升高(P<0.05或P<0.01);D组甲状腺素(T4)含量极显著升高(P<0.01)。试验第15,30天,与B组相比,C组、D组血常规、血液生化、激素各项指标均差异不显著(P>0.05)。说明在日粮中添加两种植物提取物复合物替代抗生素能改善断奶仔猪血常规和血液生化指标,提高相关激素含量,提高动物健康水平。

本试验旨在研究饲粮中添加2种植物提取物对断奶仔猪生长性能、肠道免疫功能、肠道形态结构和胰岛素样生长因子-Ⅰ(IGF-Ⅰ)含量的影响。选取胎次相近(3~6胎次)、25日龄断奶的“大白×长白”二元仔猪120头,随机分为4组,每组3个重复,每个重复10头。对照组饲喂基础饲粮,抗生素组在基础饲粮中添加100 mg/kg杆菌肽锌+20 mg/kg硫酸抗敌素,植物提取物复合物Ⅰ组在基础饲粮中添加400 mg/kg植物提取物复合物Ⅰ,植物提取物复合物Ⅱ组在基础饲粮中添加400 mg/kg植物提取物复合物Ⅱ。预试期5 d,正试期30 d。结果表明:1)与对照组相比,抗生素组、植物提取物复合物Ⅰ组和植物提取物复合物Ⅱ组的末重、平均日采食量(ADFI)、平均日增重(ADG)显著或极显著升高(P<0.05或P<0.01),腹泻率极显著降低(P<0.01)。2)与对照组相比,植物提取物复合物Ⅰ组的十二指肠和回肠绒毛高度极显著升高(P<0.01),回肠隐窝深度显著降低(P<0.05),空肠和回肠绒毛高度/隐窝深度(V/C)值显著升高(P<0.05);植物提取物复合物Ⅱ组的十二指肠、空肠和回肠绒毛高度和V/C值显著或极显著升高(P<0.05或P<0.01),回肠隐窝深度显著降低(P<0.05)。3)与对照组相比,植物提取物复合物Ⅰ组的十二指肠和空肠白细胞介素-2(IL-2)含量极显著升高(P<0.01),十二指肠和回肠白细胞介素-6(IL-6)含量极显著升高(P<0.01),十二指肠、空肠和回肠γ干扰素(IFN-γ)和分泌型免疫球蛋白A(sIgA)含量极显著升高(P<0.01);植物提取物复合物Ⅱ组的十二指肠、空肠和回肠IL-2、IL-6、IFN-γ和sIgA含量显著或极显著升高(P<0.05或P<0.01)。4)与对照组相比,抗生素组、植物提取物复合物Ⅰ组和植物提取物复合物Ⅱ组的十二指肠、空肠、回肠IGF-Ⅰ含量均极显著升高(P<0.01)。由此可见,饲粮中添加2种植物提取物复合物能够改善断奶仔猪肠道组织结构,提高肠道免疫功能,促进生长。

设空白对照组及太子参参须多糖(RPFRP)组,给药12.5、25、50、100、200、400μg/mL,分别与小鼠的脾细胞共同培养。经分光光度法筛选RPFRP对小鼠脾淋巴细胞的安全浓度范围,MTT法检测RPFRP对小鼠脾淋巴细胞增殖的影响,流式细胞术检测药物作用后小鼠脾淋巴细胞的凋亡,ELISA法检测药物作用后小鼠脾淋巴细胞上清液中细胞因子含量。结果与空白对照组相比,RPFRP可显著促进小鼠脾淋巴细胞的增殖(P<0.05),当RPFRP质量浓度在12.5μg/mL~400μg/mL时,其协同ConA可显著促进T淋巴细胞的增殖(P<0.05),当RPFRP质量浓度在50μg/mL~400μg/mL时,其协同LPS可显著促进B淋巴细胞的增殖(P<0.05);RPFRP处理组细胞总凋亡率较空白对照组明显下降(P<0.05),当RPFRP质量浓度在25μg/mL~100μg/mL时,处理组细胞的早期凋亡率和晚期凋亡率均显著降低(P<0.05),RPFRP在刺激淋巴细胞分泌细胞因子(IL-2,IL-4,IL-6和IFN-γ)中具有不同程度的促进作用(P>0.05或P<0.05)。RPFRP可促进小鼠脾淋巴细胞增殖,一定质量浓度内能减少脾淋巴细胞的凋亡,促进脾淋巴细胞因子分泌,为RPFRP的开发应用提供参考。

典型学校社区环境指示菌耐药性时空分布

Abstract :

设立福建农林大学校园内17个水域位点,进行逐月采样,分离培养大肠杆菌和肠球菌并测定其对几种常用抗生素的耐药性.结果表明,在水环境中有74.52%的分离菌存在不同程度的耐药情况.其中肠球菌对利福平耐药率最高(65.82%),对万古霉素和环丙沙星较敏感,耐药率分别为5.06%和7.59%;大肠杆菌对四环素耐药率最高(43.59%),对环丙沙星和呋喃妥因较为敏感,耐药率均为7.69%.总体而言,9—10月环境指示菌的耐药程度高于11—12月.在17个采样点中,受污水影响较强的湿地公园和观音湖地区分离的细菌耐药性更强,并且较多为多重耐药菌株.

太子参参须皂苷的免疫佐剂作用研究

本实验旨在研究太子参参须皂苷(Radix Pseudostellariae fibrous roots saponins, RPFRS)对免疫模式抗原Ovalbumin(OVA)小鼠的免疫佐剂作用。将60只雌性ICR小鼠随机分成对照空白组(CK),单独免疫OVA组(OVA),OVA+RPFRP低、中、高(50、100、200 mg/kg)剂量组(D、Z、G)。在实验的第1~5天和第16~20天,除CK组和OVA组小鼠灌胃双蒸水之外,其他组分别灌胃对应剂量的RPFRS。第6天和第21天,除CK组0.2 mL/只皮下注射PBS外,其余各组小鼠注射配OVA溶液0.2 mL/只,共免疫2次。第36天采样检测相关指标。结果表明:与OVA组相比,RPFRS组对T细胞和OVA的刺激指数均差异显著(P<0.05),D组B细胞刺激指数显著提高(P<0.05),D组和G组IL-2、IL-4含量显著升高(P<0.05),G组IFN-γ含量显著升高(P<0.05),各剂量RPFRS组OVA-IgG和OVA-IgG1含量显著提高(P<0.05),D组和G组OVA-IgG2a含量显著提高(P<0.05),D组和Z组的OVA-IgG2b含量显著提高(P<0.05),RPFRS组IL-2 mRNA、IL-4 mRNA和IFN-γ mRNA相对表达量提高(P<0.05),G组IL-10 mRNA相对表达量提高(P<0.05),RPFRS组CD3~+、CD8~+T细胞数和CD4~+/CD8~+比值显著提高(P<0.05),对CD4~+ T细胞的提升作用以D、Z组最为明显(P<0.05)。上述结果表明RPFRS能够增强小鼠免疫功能,提高OVA免疫原性,加强小鼠对OVA免疫应答能力,从而起到一定佐剂作用。

Expression profile of miRNAs involved in the hepatoprotective effects of curcumin against oxidative stress in Nile tilapia SCIE

Abstract :

Curcumin is a polyphenol with antioxidant activity that has been used to protect the health of fish livers. Our previous studies about comparative transcriptome have shown that curcumin can enhance the Nrf2-Keap1 signaling pathway and induce downstream anti-stress genes to maintain cell viability. However, the possible role of miRNAs in the protective mechanism of curcumin is not understood. In this study, the tilapia hepatocyte H2O2 stress model was used, and the miRNA expression profile for four groups (control group, curcumin group, H2O2 group, and protection group) were established by high-throughput sequencing. In our results, 278-333 types of Oreochromis niloticus miRNAs, 309-543 types of conserved miRNAs, and 535-746 types of novel miRNAs were identified in different samples. Differentially expressed miRNAs were identified by comparing miRNA expression profiles among the four groups. The expression levels were confirmed by q-PCR. The target genes of these differentially expressed miRNAs were predicted, and their functional annotations were enriched by GO and KEGG analysis, which revealed that many target genes were involved in "response to stimulus" and "antioxidant activity" in each pair of groups. Several miRNAs related to oxidative stress showed differential expression. For example, in the H2O2 group, the expression of miR-122 was decreased, and the expression of miR-21 and miR489 increased significantly. In the curcumin group, the expression of miR-153b was decreased, and the expression of miR-200a and miR-29 was increased significantly. miR-153b, miR-200a, and miR-29 may be involved in the regulation of the Nrf2-Keap1 signaling pathway by curcumin. This work might provide insights into the molecular mechanisms of miRNA regulation of curcumin on the prevention and alleviation of liver diseases in fish.

Abstract :

In recent years, the utilization of CS-MWCNT as targeted drug carriers has attracted considerable attention. Hericium erinaceus polysaccharide (HEP) has been reported as an immunostimulant to improve immune responses. This study was focussed on developing CS-MWCNT encapsulating HEP (CS-MWCNT-HEP). Using in mice peritoneal macrophages, we found the immune response could be effectively regulated by CS-MWCNT-HEP, promoted the expression of the MHCII, CD86, F4/80 and gp38. Moreover, the mice immunized with CS-MWCNT-HEP nanoparticles significantly extended PCV2-specific IgG immune response and the levels of cytokines. The results demonstrated that CS-MWCNT-HEP may be a promising drug delivery system for immuno-enhancement. (C) 2021 Elsevier B.V. All rights reserved.

Muscovy duck reovirus (MDRV) infection induces serious immunosuppression and intestinal injury in Muscovy ducklings with a high morbidity and mortality, and Astragalus polysaccharide (APS) pretreatment could efficiently protect ducklings from MDRV infection, although the underlying immunoregulatory mechanisms remain unclear. Thus, the objective of this study was to investigate effects of APS on the intestinal mucosal immunity in MDRV-infected Muscovy ducklings. A total of 190 1-day-old healthy Muscovy ducklings were randomly assigned to 3 groups (n = 50): normal control group, APS pretreatment for MDRV-infected group, and cohabitation infection group, then pretreated with 0.6 g/L APS or only drinking water followed byMDRVcohabitation infection with the remaining 40 artificially infected ducklings, respectively. At the 2, 3, 4, 6, 9 and 15 d after cohabitation infection, the intestinal samples were prepared to measure intestinal parameters including villus length, villus length/crypt depth (V/C) ratio, and wall thickness, together with counts of intraepithelial lymphocyte (IEL) and goblet cell (GC) by hematoxylin-eosin staining. Meanwhile, ileal secretory IgA (sIgA) and duodenal cytokine levels of IL-4, IL-6, IL-15, tumor necrosis factor-alpha, and interferon gamma were detected by the ELISA and radioimmunoassay, respectively. The results showed that APS significantly improved intestinal injuries of villi length, V/C ratio, and wall thickness of the small intestine infected with MDRV, effectively inhibited the reduction of IEL and GC caused by MDRV infection, subsequently increased sIgA and all the cytokine secretions at most time points, suggesting that APS pretreatment can effectively stimulate mucosal immune function by improving intestinal morphology and repair MDRV caused injures of small intestinal mucosal immune barrier in infected ducklings. Our findings lay the foundation for further application of APS in prevention and treatment of MDRV infection.

Abstract :

In this study, the enzyme degradation of Hericium erinaceus polysaccharide (HEP) was successfully modified with endo-rhamnosidase to obtain the enzymatic hydrolysis of Hencium erinaceus polysaccharide product (EHEP). The gas chromatography-mass spectrometry (GC-MS), high performance gel permeation chromatography ( HPGPC), Fourier transformed infrared spectrometry (FT-IR), scanning electron microscopy (SEM), atomic particle microscopy (AFM), nuclear magnetic resonance (NMR) and particle size distribution were used to characterize polysaccharides. In vitro, EHEP significantly enhanced the phagocylosis, NO, CD40 and CD86 by macrophage than HEP. In vivo, female Balb/c mice were injected respectively with EHEP and HEP after administrated with cyclophosphamide, once a day for 7 days. On days 11, the morphology and structure of jejunal sections, immunofluorescence of spleen and peritoneal macrophages were determined. These results indicated that the enzymatic hydrolysis product could enhance the activation of peritoneal macrophages, and enhance the immunomodulation function of HEP. This study demonstrated that enzymatic modification was an effective method to improve the activities of HEP, and could be developed as a potential technology for use in pharmaceutical and cosmeceutical industry.
