Plant morphological and functional characteristics combined with elevation and aspect influence phytogenic mound parameters in a dry-hot valley SCIE

Abstract :

Isolated plants growing on steep slopes with erodible bedrock are usually associated with the formation of phytogenic mounds in semiarid and arid ecosystems. Identifying the plant characteristics that affect phytogenic mounds and predicting a plant community's potential for erosion control are of great interest to workers seeking to improve the management of degraded ecosystems. We studied the phytogenic mounds of three dominant perennial grass species found between 1,400 and 1,700 m above sea level in Jiangjiagou Gully: Heteropogon conforms, Eulaliopsis hinata, and Eriophorum comosum. Morphological and functional characteristics were measured for a total of 42 individuals from the three grass species on both sunny and shady slope aspects. Soil samples were collected from the mounds and from bare land 30 cm upslope from each mound to determine particle size distribution. Morphological and functional characteristics, as well as mound parameters, were summarized using principal component analysis (PCA). Pearson's correlation analysis, linear regression analysis, and redundancy analysis were conducted to identify the relationships between plant characteristics and mound parameters. Then we used analysis of covariance to examine the combined effects of aspect and elevation and plant characteristics on mound parameters.We found that there are no differences in morphological characteristics, functional characteristics, or mound parameters between the three grass species, or soil particle size distribution between mounds and bare land. PCA shows that mound parameters are best correlated (positively) with interception length and total dry weight. Crown length in the direction of the slope and root width are also positively correlated with maximum height and maximum width but to a lesser extent. Plant morphological and functional characteristics can explain greater than 60% of the variance of all mound parameters except maximum length, with the greatest contributions from crown length and interception length. Aspect and elevation, combined with some plant characteristics, also affected mound parameters. Our study indicates that perennial grass species can accumulate sediment deposits during growth in a dry-hot valley. Our results provide useful information for evaluating the function of plants in erosion control and for identifying ways to improve the effectiveness of species in mound formation in dry-hot valley ecosystems.

Abstract :

As one of the most popular Chinese tea products, oolong tea involves the most complicated manufacturing process and the most abundant tea germplasm resources. However, the unclear genetic backgrounds of oolong tea germplasms have been biological limitations for tea breeding and quality control. In this study, high-throughput SNP technology was used to analyze the genetic diversity of 100 oolong tea landraces and cultivars. Ninety-six pairs of primers were selected and validated from the expressed sequence tag (EST) database of Camellia sinensis. The results verified 75 SNP loci that are able to accurately and efficiently determine the genetic relationships among Chinese oolong tea germplasms. The DNA fingerprints of 4 main oolong tea-producing areas were constructed by tracing the information of each site of the SNP. The Guangdong oolong tea germplasms were a relatively distinct group, while the genotypes of Taiwan oolong tea were similar to those of populations in Fujian. The tea germplasms in Southern and Northern Fujian had the largest intersections, and they also showed their own characteristics. Our results provide guidance for the identification, integration, and selection of parents for tea plant germplasms. Moreover, our study provides an effective tool to protect the diversity of tea germplasms and to assist in future breeding work.

Although parasites and microbial pathogens are both detrimental to insects, little information is currently available on the mechanism involved in how parasitized hosts balance their immune responses to defend against microbial infections. We addressed this in the present study by comparing the immune response between unparasitized and parasitized pupae of the chrysomelid beetle, Octodonta nipae (Maulik), to Escherichia coli invasion. In an in vivo survival assay, a markedly reduced number of E. coli colony-forming units per microliter was detected in parasitized pupae at 12 and 24 h post-parasitism, together with decreased phagocytosis and enhanced bactericidal activity at 12 h post-parasitism. The effects that parasitism had on the mRNA expression level of selected antimicrobial peptides (AMPs) of O. nipae pupae showed that nearly all transcripts of AMPs examined were highly upregulated during the early and late parasitism stages except defensin 2B, whose mRNA expression level was downregulated at 24 h post-parasitism. Further elucidation on the main maternal fluids responsible for alteration of the primary immune response against E. coli showed that ovarian fluid increased phagocytosis at 48 h post-injection. These results indicated that the enhanced degradation of E. coli in parasitized pupae resulted mainly from the elevated bactericidal activity without observing the increased transcripts of target AMPs. This study contributes to a better understanding of the mechanisms involved in the immune responses of a parasitized host to bacterial infections.

Fresh jasmine flowers have been used to make jasmine teas in China, but there has been no complete information about volatile organic compound emissions in relation to flower developmental stages and no science-based knowledge about which floral stage should be used for the infusion. This study monitored volatile organic compounds emitted from living flowers of Jasminum sambac (L.) Ait. 'Bifoliatum' at five developmental stages and also from excised flowers. Among the compounds identified, alpha-farnesene, linalool, and benzyl acetate were most abundant. Since alpha-farnesene is synthesized through the Mevalonate pathway, four genes encoding 3-hydroxy-3-methylglutaryl coenzyme A synthase, 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR), farnesyl pyrophosphate synthase, and terpene synthase were isolated. Their expression patterns in living flowers at the five stages and in excised flowers coincided with the emission patterns of alpha-farnesene. Application of lovastatin, a HMGR inhibitor, significantly reduced the expression of the genes and greatly decreased the emission of alpha-farnesene. The sweet scent was diminished from lovastatin-treated flowers as well. These results indicate that alpha-farnesene is an important compound emitted from jasmine flowers, and its emission patterns suggest that flowers at the opening stage or flower buds 8 h after excision should be used for the infusion of tea leaves.

Abstract :

Because immune defenses are often costly employed, insect immunocompetence cannot be always maintained at its maximum level. Here, the oriental fruit fly, Bactrocera dorsalis (Hendel), was used as a study object to investigate how its immune defenses varied with the developmental stage change and mating access. Our data indicated that both phenoloxidase (PO) activity and antibacterial activity significantly increased from new larvae to pupae but decreased in adults after emergence. Furthermore, both the PO activity and antibacterial activity in the hemolymph of copulated male and female adults were dramatically higher than that of virgin male and female ones, respectively. It provided the evidence that copulation could increase the magnitude of immune defense in hemolymph of B. dorsalis. Together, these results suggest that B. dorsalis possess a flexible investment strategy in immunity to meet its specific needs based on the endo-and exogenous factors, such as their distinct food source and living environments.

Abstract :

背景:近10年来干细胞研究所取得的巨大进展,不仅影响和促进了生物学及其相关基础科学,而且还在医学、药物开发、农业等许多领域得到广泛的应用,目前已成为研究的热点问题。目的:在干细胞的定义上还存在一些需要探讨的问题,明确定义将有利于干细胞研究的快速发展。方法:回顾干细胞的发展和概念提出,特别是通过中英文权威定义的比较,提出作者的看法。结果与结论:干细胞的定义上还存在一些问题值得探讨,特别是一些中英文表述不同影响了理解。例如,"多能干细胞"对应译成两个单词:Pluripotent Stem Cell和Multipotent Stem Cell,然而Pluripotent和Multipotent两个单词的英文意义不同。为了学术交流和沟通,作者提出将Pluripotent Stem Cell称为"万能干细胞",而保留Multipotent Stem Cell为"多能干细胞"的定义。以上结论供广大同仁探讨,以利于抛砖引玉。

Abstract :

背景:诱导多能干细胞可以绕过胚胎干细胞存在的伦理学问题,已成为目前干细胞研究的热点问题。目的:探讨诱导多能干细胞的研究进展以及需要解决的问题。方法:回顾分析诱导多能干细胞的发现,近年来的研究过程,目前存在的问题。检索汤森路透WebofScience数据库关于诱导多能干细胞和p53基因研究相关的文献进行分析。结果与结论:近年来国内外学者对诱导多能干细胞进行大量研究。例如,日本正在筹备建立干细胞库,将为视网膜等疾病治疗提供基础。但是,在诱导多能干细胞能广泛应用之前,还面临许多安全问题,其中p53相关的功能性研究是必不可少的紧迫问题,p53基因中的其他成员p73也参与诱导多能干细胞的产生和分化也需要进一步深入。关于p53功能的发现将伴随着干细胞研究,促进再生医学的深入和发展。
