Abstract :
In this study, spent mushroom substrates (SMSs) were fermented anaerobically at room temperature to gain liquid SMSs (LSMSs) that were used to remove nitrogen from the piggery wastewater with a low C/N ratio in a sequencing batch reactor (SBR) and solid SMSs (SSMSs) that were utilized to adsorb Pb2+ from Pb2+-containing wastewater in a fixed-bed reactor (FBR). After LSMSs supplement, the removal efficiency of both total nitrogen (TN) and NH4+-N increased from around 50% to 60-80%. High-throughput sequencing results presented an obvious change in microbial diversity, and some functional microorganisms like Zoogloea and Hydrogenophaga predominated to promote nitrogen removal. Pb2+ did not emerge from the effluent until 240 min with the corresponding concentration being less than 3 mg/L when using 30-day SSMSs as adsorbents, and it was demonstrated to be appropriate to use the Thomas model to predict Pb2+ sorption on SSMSs. Although various functional groups played a role in binding ions, the carboxyl group was proved to contribute most to Pb2+ adsorption. These results certified that the anaerobically fermented SMSs are decidedly suitable for wastewater treatment.

Subaqueous wetland sediments (SWSs) and banana peels (BPs), respectively used as anaerobic inoculums and raw materials, were valorized for electricity generation in membrane-less and biocathode-containing microbial fuel cells (MLBC-MFCs). The maximum current density was 78.2 mA/m(2) for banana peel extracts (BPE) and 91.3 mA/m(2) for anaerobically fermented banana peel extracts (ABPE). The polarization data indicated that Ohmic resistances were high that were 3.5, 4.2 and 9.3 k Omega when MLBC-MFCs were fed with ABPE, BPE and sodium acetate (SA), respectively. The ultraviolet-visible absorption spectroscopy coupled with three-dimensional fluorescence spectroscopy was used to explore the mechanisms of generating electricity. Results showed that SWSs provided functional microorganisms and a source of organic matter, while BPs were more helpful for the growth of microorganisms than SA and the microorganisms attached on the cathode primarily functioned as biocatalysts. Overall, this study highlights the simplicity and scalability of MLBC-MFCs equipped with SWSs and BPs.

Acinetobacter sp. TX5 immobilized with spent Hypsizygus marmoreus substrate (SHMS) was used to treat the raw piggery wastewater (RPW). In batch experiments, NH4+ -N in the diluted RPW decreased from initial 34.95 mg/L to 3.83 mg/L at 8 h with the removal efficiency (RE) being 89%, and the beads immobilized with SHMS were comparable to those immobilized with activated carbon. In continuous experiments, the RE ranged from 74% to 95% for NH4+ -N, from 73% to 93% for TN and from 54% to 82% for COD when the RPW was treated in a fixed-bed reactor packed with SHMS-immobilized TX5. The isotope analysis and enzyme purification indicated simultaneous nitrification and denitrification existing in TX5. This is the first time that spent mushroom substrates have been used to immobilize Acinetobacter species to treat the real RPW and a denitrifying nitrite reductase (dNiR) has been purified to make the nitrogen removal pathway in this species clearer. The spent compost of Hypsizygus marmoreus (SCHM) modified by NaOH solution was used as a novel biosorbent (MSCHM) to remove Pb(II) ions from aqueous solution in this study. Batch adsorption experiments revealed that the uptake capacity increased from 54.8 mg/g by SCHM to 72.5 mg/g by MSCHM and the adsorption efficiency improved regardless of the initial Pb(II) concentration. In the fixed-bed column experiments, the breakthrough and exhaustion time were extended/shorten about 2-11 times with the flow rate, bed depth and initial concentration changing from 20 mL/min, 12 cm and 45 mg/L to 40 mL/min, 28 cm and 145 mg/L, respectively. The adsorption process was predicted and described well by Thomas model indicating the external and internal diffusions were not the bottleneck of the process. The regeneration experiments were conducted to validate the reuse effectiveness of MSCHM, and the final regeneration efficiency achieved to 73%. Furthermore, the preliminary adsorption mechanisms of Pb(II) onto MSCHM were investigated by Brunauer-Emmett-Teller method, scanning electron microscope and Fourier transform infrared (FT-IR) spectrometer, and carboxyl, hydroxyl and amide might be important functional groups adsorbing Pb(II) ions.

The membrane-less and biocathode-containing microbial fuel cells (MLBC-MFCs) were used for the electrical energy production from different wetlands, and the corresponding mechanisms were explored based on the characteristics of dissolved organic matters (DOM) in this study. The average voltage output on indoor MLBC-MFCs was 60 mV for the saltmarsh wetland water (SWW), 48 mV for the forest wetland water (FWW) and 71 mV for the FWW supplemented with sodium chloride (FWWS). For the field experiments, as high as 800 mV was obtained from the MLBC-MFCs installed in the forest wetland and the voltage output varying between 200 mV and 800 mV maintained approximately for half a year except for a few days with an extremely low temperature, while the voltage output from the MLBC-MFCs installed in the saltmarsh wetland went down to below 10 mV within 37 days likely due to the fide. The DOM before and after MFCs treatment was analyzed using the ultraviolet-visible absorption spectroscopy (UV-Vis) and the fluorescence excitation-emission matrix (EEM). Results showed that both carbon sources and anaerobic microorganisms included in subaqueous wetland sediments (SWSs) were vital for the electricity generation, and the food used to produce electrical energy mainly came from SWSs instead of wetlands water.

(3S)-Acetoin and (2S,3S)-2,3-butanediol are important platform chemicals widely applied in the asymmetric synthesis of valuable chiral chemicals. However, their production by fermentative methods is difficult to perform. This study aimed to develop a whole-cell biocatalysis strategy for the production of (3S)-acetoin and (2S,3S)-2,3-butanediol from meso-2,3-butanediol. First, E. coli co-expressing (2R,3R)-2,3-butanediol dehydrogenase, NADH oxidase and Vitreoscilla hemoglobin was developed for (3S)-acetoin production from meso-2,3-butanediol. Maximum (3S)-acetoin concentration of 72.38 g/L with the stereoisomeric purity of 94.65% was achieved at 24 h under optimal conditions. Subsequently, we developed another biocatalyst co-expressing (2S,3S)-2,3-butanediol dehydrogenase and formate dehydrogenase for (2S,3S)-2,3-butanediol production from (3S)-acetoin. Synchronous catalysis together with two biocatalysts afforded 38.41 g/L of (2S,3S)-butanediol with stereoisomeric purity of 98.03% from 40 g/L meso-2,3-butanediol. These results exhibited the potential for (3S)-acetoin and (2S,3S)-butanediol production from meso-2,3-butanediol as a substrate via whole-cell biocatalysis.

为了从低碳氮比的废水中有效地除氮,在序批式反应器中研究了利用废弃载培料水解物作为反硝化碳源的可行性;并进行了功能微生物的分离纯化,以进一步探讨反应器脱氮的机理。结果表明,废弃载培料的添加,在30 d的运行过程中序批式反应器的性能明显地得到改善。TN的去除率从46.9%提高到69.7%~93.0%,添加不同的废弃载培料到序批式反应器中没有负面影响。共筛选分离到14株好氧反硝化细菌,其中假单胞菌属4株,不动杆菌属8株,肠杆菌属1株,未知菌株1株。因此,废弃载培料是一种非常有前途的脱氮碳源,具有成本低、效率高的特点。

In order to remove nitrogen effectively from the wastewater with a low C/N ratio, the feasibility of using spent mushroom compost (SMC) hydrolysates as carbon sources for denitrification was investigated in a sequencing batch reactor (SBR). With SMCs supplement, the SBR performance was improved obviously within the 180 days of operation. The total nitrogen removal was promoted from 46.9% to 81-89.4%, and no negative impact induced by different SMCs on the SBR system was observed. The abundance of functional genes including amoA, nirS/K, norB and nosZ in the active sludge was quantified by qPCR, and most of them elevated after SMC was fed. 16S rRNA gene high-throughput sequencing showed that the significant change in microbial community not only promoted pollutants removal but also benefited the stability of the reactor. Therefore, SMC could be an extremely promising carbon source used for nitrogen removal due to its cost-effective and efficient characteristics.

This work evaluates the efficiency of ammonium removal from piggery wastewater using a sequencing batch reactor (SBR) inoculated with a newly isolated strain of Pseudomonas putida HJH1. The strain HJH1 not only could survive and remove up to 72.4 mg/L NO2--N under aerobic conditions, but it also has good performance for simultaneous nitrification and denitrification (SND) with no nitrite accumulation. The SBR system was able to consistently remove: 1) 76.4-100% ammonium nitrogen and 71.4-100% COD from artificial wastewater in stage 1, and 2) 89.2-99.1% ammonium nitrogen and 82.4-100% COD from piggery wastewater in stage 2. During the whole operation, the strain HJH1 predominated in the SBR all the time to function together with other bacteria. Results indicated that the SBR system inoculated with Pseudomonas putida HJH1 can efficiently remove ammonium nitrogen from piggery wastewater, thereby having potential applications for future nitrogen removal.
